Changes in Lipid Composition and Peroxidation by Insulin and Aminoguanidine in STZ-induced Rat.

Abstract
In the streptozotocin-induced diabetic rats were examined the changes of glucose concentration, lipid composition in blood and the levels of lipid peroxidation of RBC ghost and organelles in hepatocyte as well as the activities of glutathione peroxidase in response to the peroxidation As expected, blood glucose concentration of the diabetic group showed a high significant increase in comparison to the control. In contrast, the experimental groups treated with insulin or aminoguanidine (AG) showed both the hypoglycemic effects. Tests on the lipid composition in plasma of the diabetic group, were also resulted in the higher cholesterol and triglyceride concentrations than the control group. The treatment with insulin or AG didn¡¯t show a significant decreasing effect on the lipid concentration in plasma but the blood cholesterol was significantly lower in the AG-group than the diabetic in the oxidation tests, RBC-ghost, mitochondria and microsome of the diabetic rat were oxidized at a much higher rate, but the rate was retarded by insulin or AG-treatment. In accordance with its higher oxidation rate, the diabetic group also showed an overall higher glutathione peroxidase activity in mitochondria and microsome. In conclusion, the chemically induced diabetes mellitus caused not only the change of lipid components in plasma, but also the higher oxidative tendency. This higher tendency may be due to the already reported autooxidation in process of mallard reaction between glucose and protein in plasma. The treatment with insulin or AG to suppress the diabetic condition was each resulted in positive effects showing similar values to those of the control group. Although more precise experiments are needed to explore the effects of AG-treatment for diabetes mellitus, the AG showed a good possibility to prevent the complications of the diabetes mellitus through its antioxidant effect.
